The same probe was used to confirm hybrid nature of somatic fusion plants of D. catholica + B. juncea, B. spinescens +B. juncea, T. ballii +B. campestris (Kirti et al. unpublished) and somatic hybrids between C. sativa + B. carinta (Narasimhulu et al. unpublished). Molecular analysis of B. carinata synthesized by protoplast fusion of parental species was made by probing with a B. juncea derived genomic clone BJG 425 (Mohaptra 1991). The clone binds differentially to Hind III fragments of the two diploid parental species.
Ballii+B. juncea all other hybrid combinations showed mitochondria predominantly from the cultivated parent. Mixed mitochondrial DNA sequences of both parental species were observed in somatic hybrids B. Juncea(Kirti et al unpublished). juncea. Approximately 28% of all hybrid analysed, and more than 50% of the hybrids in these combinations, showed mitochondrial rearrangements (Table 4). Protoplast fusion thus provides a unique oIJportunity of recovering novel mitochondrial gene combinations which could not have been possible by conventional hybridisation.
